This material was designed to provide a detailed review of the preparation of spleen cells for immunophenotyping. Speen must be reduced to a single cell suspension before cells can be stained with monoclonal antibodies and then run on the flow cytometer for determination of T cell, B cell subsets as well as other cell enumeration procedures.
